I am going to Canada in January and want to travel from Calgary to Vancouver and very much want to see Banff, Lake Louise. What is the best way of doing this, and the best places to stay?

Driving is best but could be dangerous due to snow. Best place to stay. Chateau Lake Louise, Expensive but worth it.
Check out viarail.ca, and another company called Canadian Rockies by Rail. You can stay right on the train if you like, but depending on your age/style, there’s everything from hostels to Grand hotels like the Banff Springs Resort, or the very Grand Chateau Lake Louise. It will all take your breath away, and hopefully in January there is enough snow for skiing/snowboarding. If you have a limited budget you can rent a car and wander safely on your own.

If you can, overshoot to Vancouver Island and Victoria.
Before you go north or West, you might want to drop down into Montana for a few days.
Go by one-way Hire Car.
I am not sure if you want to drive or not but Calgary is closer to Banff than Lake Louise so if you are flying in and out of Calgary I would suggest going to Banff first via Highway 93 and do a loop up through the Rockies and then go trough Edmonton back to Calgary.

The best way to travel is by train, as for the best places that is a two edged sword! What I like you may not and vice versa so I will not presume to answer that part of the question, sorry. But I do recommend the train, that way you get to see the real Canada and not just the bits you can see from a road, like going by bus or car, you are actually IN the scenery, and very nice scenery it is, you will love it! I did! Hope you have a great time!
I’d take a plane myself, but if you want to go by road take the Greyhound. They make several stops. The roads through the mountains can be brutal in the wintertime especially if you’re not familiar with driving under those conditions.
